We aim to identify target sequences of suz12, a member of the polycomb group proteins (PcG) PRC2 (polycomb repressive complex 2). We study the role played by this polycomb complex on genetic plasticity during embryonic development in undifferentiated mouse embryonic stem cells (day 8). Suz12 is required for trimethylation of the tail of histone H3 on lysine 27 (H3K27me3). This epigenetic modification of chromatin induces dynamic repression of many target genes involved in developmental pathways. The experiment reported here aimed to set up the ChIP-on-chip procedure in our lab, and this study was already published by the Young RA group (Boyer LA and al., Nature (2006), 441, 349-353).
