Biliary tract cancers are rare cancers with a poor prognosis. Long lacking a standard of care, second-line treatment of advanced biliary tract cancers has seen the establishment of FOLFOX as a standard, as well as the advent of targeted therapies, underlining the importance of molecular profiling at the earliest possible stage of management. This mini-review provides an update on the evolution of second-line management of advanced biliary tract cancers in the era of personalized medicine.
