T cell checkpoint blockade therapies have revolutionized treatment protocols and prognosis in patients with cancer. Pointed out by the success of PD-1 plus CTLA-4 blockade in patients with melanoma, the perspective of new synergistic immunotherapy combinations seems to be an important opportunity to improve outcomes for patients. We will first focus on a review of immunotherapy combinations that are approved in solid tumors and emerging targets with reported pre-clinical efficacy which are currently evaluated through ongoing clinical trials. The main limit of immunotherapy combination being the occurrence of immune-related adverse events that can be severe and challenging to manage, we will then present RETROICITOX results, a retrospective multicentric study of patients treated with Nivolumab and Ipilimumab with the aim to identify predictive factors of immune-related adverse events.
