Various fluorescent labeled prooligonucleotides bearing a variable number of tBu-SATE phosphotriester and phosphodiester linkages, and exhibiting various lipophilic behaviors were synthesized on solid support. Among them, those with relative high lipophilicity were shown to be rapidly and efficiently internalized into HeLa cells. Fluorescence patterns of internalization were consistent with a diffusion mechanism resulting in the appearance of uniform cytoplasmic distribution and a nuclear accumulation as confirmed by confocal microscopy.
