INTRODUCTION: Obesity is an important risk factor for chronic kidney disease (CKD). It carries hemodynamic and morphologic changes in the kidney resulting in renal function reduction. CKD is associated to glomerulosclerosis and tubulointerstitial fibrosis and involves collagen deposition. The aim of the study was to evaluate the impact of obesity on kidney fibrosis in a « cafeteria diet » and 5/6 nephrectomy rat model.METHODS: Twenty four rats were fed with a « cafeteria diet » and 10 rats with a standard diet. Half of the rats underwent 5/6 nephrectomy at 4 months. Expression of collagens types 1, 3 and 4 was quantified by immunohistochemistry at 6 months.RESULTS: Cafeteria diet – rats were hyperglycemic and hypertensive. Their weight was higher than that of the rats fed with standard diet. Nephrectomized rats had increased serum creatinine levels, which were even higher in the « cafeteria » group (130±66 µM) compared to standard diet group (57±7µM)(p<0.05). Serum creatinine level was comparable in non nephrectomized animals, 31.7±1.2µM for the cafeteria group and 34.2±5.1 µM for standard diet group. Expression of collagens 1, 3 and 4 were increased in both groups of nephrectomized rats with no clear difference between them in the analysed rats (preliminary results).CONCLUSIONS: Preliminary results suggest an influence of obesity on renal function. Collagen expression is increased in both nephrectomized 5/6 groups but is not increased in the obese model suggesting that obesity-related renal dysfunction is unrelated to fibrosis.
